ORCA · TWIST WGS LIBRARY PREP
Orca automates the Twist hybrid capture workflow from library preparation and cleanup through hybridization, bead-based capture, post-capture amplification, and library quantification.
Key benefits
Eliminate hands-on pipetting across fragmentation, end repair, ligation, and cleanup
No inter-operator hand-offs
Less than 30 minutes of hands-on time

How the Twist workflow runs on Orca
Orca automates the Twist Library Preparation EF Kit 2.0 chemistry end-to-end: enzymatic fragmentation, end repair/A-tailing, adapter ligation, and bead-based cleanup, finishing with amplification-ready WGS libraries.
Built-in bead cleanup steps replace manual AMPure handling at every stage, so library quality stops depending on who’s running the deck.
Day 1
Fragmentation, end prep/A-tailing
Adapter ligation
Library amplification
Post-amplification clean up
Quantification, normalization, pooling
Bead-based drying (Twist Dry Down Beads)
Hybridization (overnight)
Minutes total: 145 min, then overnight hybridization
Day 2
Streptavidin bead prep + wash
Post-capture amplification

Load consumables
With under 30 minutes of hands-on setup, Orca takes genomic DNA straight through to indexed, sequence-ready WGS libraries, same-day or overnight, depending on your run schedule.

Performance data
Based on a matched technical study: Twist WGS libraries prepared from one genomic DNA sample as two technical replicates (A1, D1) on Orca vs. manual workflow, using the Twist Library Preparation EF Kit 2.0. Libraries were pooled equimolarly and sequenced at Strand Life Sciences, 3 Gb requested per library.
Sequencing output matched manual prep
Figure A. Estimated paired-end sequence output (Gb) vs. the requested 3 Gb per library. Orca-prepared libraries generated 4.9–5.3 Gb of data per library, closely matching manual preps and exceeding the requested low-pass allocation.
High-quality reads across the full read length

Figure B. Mean per-base Phred quality for all R1 and R2 FASTQ files. Scores remained within the high-quality range across the full read length for all eight FASTQ files, with a gradual decline toward the 3’ end consistent with the shared sequencing run rather than a preparation-specific effect. Per-base sequence quality passed across all eight FASTQ files and remained above Q30 throughout 151-bp paired-end reads.
